Analysis of bacteriostatic substances baicalein and glycyrrhizin in Niuhuang Jiedu tablets by high performance liquid chromatography-tube dish method

摘要：联合运用高效液相色谱法和管碟法分析牛黄解毒片中抑菌物质黄芩苷 、甘草苷。 以 Diamonsil C18柱（250 mm×4.6 mm，5 μm）为分析柱，0.15% 磷酸水溶液为流动相 A、乙腈为流动相 B，流量为 1.0 mL/min，检测波长为 280 nm，柱温为 30 ℃，进样体积为 10 μL。采用高效液相色谱法测定 12 批不同厂家的牛黄解毒片，记录色谱图；运用《中药色谱指纹图谱相似度评价系统》（2012 年版）建立指纹图谱，并进行相似度评价和共有峰指认。采用管碟法，以阿莫西林为对照，测定牛黄解毒片对金黄色葡萄球菌的抑菌活性；使用 SPSS 21.0 软件对牛黄解毒片的抑菌活性指标值与指认峰峰面积进行灰色关联度分析。建立高效液相色谱指纹图谱，12 批不同厂家牛黄解毒片的相似度均不低于 0.937，并确认甘草苷与黄芩苷为共有峰；甘草苷的灰色关联度值为 0.696，黄芩苷的灰色关联度值为 0.680。黄芩苷与甘草苷是牛黄解毒片中的 2 种抑菌物质。
Abstract：High performance liquid chromatography was combined with tube dish method to analyze the bacteriostatic substances baicalin and glycyrrhizin in Niuhuang Jiedu tablets. Diamonsil C18 column (250 mm×4.6 mm, 5 μm) was used as the analysis column, 0.15% phosphoric acid aqueous solution was mobile phase A, acetonitrile was mobile phase B, the flow rate was 1.0 mL/min, the detection wavelength was 280 nm, the column temperature was 30 ℃, and the injection volume was 10 μL. 12 batches of Niuhuang Jiedu tablets from different manufacturers were determined by high performance liquid chromatography, and the chromatograms were recorded. The "Chinese Medicine Chromatography Fingerprint Similarity Evaluation System" (2012 edition) was used to establish the fingerprint, and the similarity evaluation and common peak identification were carried out. The antibacterial activity of Niuhuang Jiedu tablets against staphylococcus aureus was determined by tube dish method, with Amoxicillin as the control. SPSS 21.0 software was used to analyze the grey correlation between the antibacterial activity index value of Niuhuang Jiedu tablets and the designated peak area. The high performance liquid chromatography fingerprint was established, and the similarity of 12 batches of Niuhuang Jiedu tablets of different manufacturers were not less than 0.937, and it was confirmed that glycyrrhizin and baicalin were common peaks, the gray correlation value of glycyrrhizin was 0.696, and the gray correlation value of baicalin was 0.680. Baicalin and glycyrrhizin were two bacteriostatic substances in Niuhuang Jiedu tablets.
