麻杏止咳糖浆的薄层色谱法鉴别及高效液相色谱法测定其中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷

摘要:采用薄层色谱法鉴别麻杏止咳糖浆中甘草药材,用高效液相色谱同时测定麻杏止咳糖浆中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷的含量。以乙腈-0.02 mol/L磷酸溶液(含0.2%的三乙胺,用磷酸调节pH值至3.0) (体积比为5∶95)为流动相,流量为1.0 mL/min ,柱温为30 ℃,检测波长为210 nm。薄层色谱中可见甘草的特征,盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷进样质量分别在1.056~6.336 μg、1.003~6.018 μg 和0.889~5.334 μg范围内与色谱峰面积线性关系良好,相关系数分别为0.999 9、0.999 6、0.999 2,测定结果的相对标准偏差分别为1.5%、1.8%和1.6%(n=6),平均加标回收率分别为100.7%、98.5%和104.4%。该方法操作简便、重现性好,可用于完善麻杏止咳糖浆的质量标准。
Abstract:Glycyrrhiza uralensis was identified by TLC, and the contents of ephedrine hydrochloride, pseudoephedrine hydrochloride and amygdalin in Maxing Zhike syrup were determined by using HPLC. The mobile phase was acetonitrile-0.02 mol/L phosphoric acid solution (containing 0.2% triethylamine, pH was adjusted to 3.0 with phosphoric acid)(volume ration was 5∶95), the flow rate was 1.0 mL/min, the column temperature was 30 ℃, and the detection wavelength was 210 nm. The characteristic chromatogram of Glycyrrhiza uralensis could be detected in TLC chromatogram. Sample mass of ephedrine hydrochloride, pseudoephedrine hydrochloride and amygdalin had good linear relationship with the chromatopgraphic peak area in the range of 1.056-6.336 μg, 1.003-6.018 μg and 0.889-5.334 μg, respectively with the correlation coefficients of 0.999 9, 0.999 6, and 0.999 2. The relative standard deviations of the determination results were1.5%,1.8% and 1.6% (n=6), and the average recoveries were 100.7%, 98.5% and 104.4%, respectively. The established method is simple, reproducible and it can be used to improve the quality standard of maxing Zhike syrup.
