高效液相色谱法测定北沙参中的蔗糖及其指纹图谱

摘要:采用高效液相色谱(HPLC)法测定北沙参中的蔗糖含量,建立北沙参的HPLC指纹图谱。以Innoval NH2 色谱柱(250 mm×4.6 mm,5 洀)为分离柱,检测器为Waters 2420蒸发光散射检测器,以乙腈-水(80∶20)为流动相,漂移管温度为60 ℃,喷雾器加热级别为100%,气体压力为137.90 kPa,对北沙参样品溶液进行指纹图谱测定和蔗糖含量测定。蔗糖的质量在7.30~72.95 最范围内与色谱峰面积线性关系良好,相关系数为r=0.9997。样品加标回收率为98.85%,测定结果的相对标准偏差小于3%(n=6)。指纹图谱测定研究中确定了6个共有峰,药材样品与对照指纹图谱的平均相似度为0.9849,10批药材样品间的相似度为0.917~0.999。建立的指纹图谱测定方法能反应不同来源北沙参药材之间的差异,可作为北沙参药材质量控制的有效手段。 
Abstract:The high performance liquid chromatography(HPLC) method was used to determine the sucrose content in Glehniae Radix, and the HPLC fingerprint of Glehniae Radix was established. The Innoval NH2 column (250 mm×4.6 mm, 5 μm) was used as the separation column; the detector was Waters 2420 evaporative light scattering detector(ELSD); acetonitrile-water (80∶20) was used as mobile phase; drift tube temperature was 60 ℃; sprayer heating level was 100%; gas pressure was 137.90 kPa. Glehniae Radix sample solution of fingerprint and sucrose content were determined. The mass of sucrose had a good linear relationship with the peak area in the range of 7.30-72.95 最, and the correlation coefficient was r=0.9997. The sample recovery rate was 98.85%, and the relative standard deviation of the measurement result was less than 3%(n=6). Six common peaks were identified in the fingerprint measurement study, the average similarity between the medicinal material samples and the control fingerprint was 0.9849, and the similarity between 10 batches of medicinal material samples was 0.917-0.999. The established fingerprint determination method can reflect the differences between different sources of Glehniae Radix medicinal materials, and it can be used as an effective method for the quality control of Glehniae Radix medicinal materials.
