高效液相色谱法测定四季青中长梗冬青苷

摘要:  建立了高效液相色谱法测定四季青中长梗冬青苷含量的方法。采用C18色谱柱,蒸发光散射检测器,以甲醇-异丙醇(9∶1)与水-异丙醇(9∶1)为流动相梯度洗 脱,流量为1.0 mL/min,柱温为30℃,气体流量为2.5 L/min, 漂移管温度为50 ℃。长梗冬青苷的质量在0.16~16 μg范围内线性关系良好,线性相关系数为0.999?9, 检出限为0.016 mg/mL。样品的加标回收率为96.25%~98.89%,测定结果的相对标准偏差为0.30%~3.56% (n=6)。该方法简便、准确、灵敏度高、重现性好,且 满足药典要求,可用于四季青中长梗冬青苷的含量测定。  
Abstract:The high performance liquid chromatography method was established to determine Pedunculoside in Ilex chinensis Sims. The C18 column (4.6 mm ×250 mm, 5 μm) and ELSD were used, gradient elution was performed with methanol-isopropyl alcohol (9∶1) and water-isopropyl alcohol (9∶1) as mobile phase with flow rate of 1 mL/min, the column temperature was 30 ℃, the temperature of detector was 50 ℃, the flow rate of N2 was 2.5 L/ min. The linear range of Pedunculoside was 0.16-16 μg with the linear correlation coefficient of 0.999?9, the detection limit was 0.016 mg/mL. The recoveries of standard addition were 96.25%-98.89%, and the relative standard deviations of determination results were 0.30%-3.56% (n=6). The method is simple, accurate, sensibility, reproducible and meet pharmacopoeia requirements. It can be used for the determination of Pedunculoside in Ilex Chinensis Sims.
