离子色谱法同时测定尿液中草酸和硫氰酸盐

摘要:建立抑制电导检测-离子色谱法同时测定尿样中草酸和硫氰酸盐的方法。通过试验优化确定了各项色谱条件,以SH-AC-3 型阴离子交换柱为分离柱,以10.0 mmol/L Na2CO3 为淋洗液,流量为1.0 mL/min,等度洗脱可将草酸和硫氰酸盐与尿液中大量共存的氯化物、磷酸盐和硫酸盐等常见阴离子完全分离,采用抑制电导检测。草酸和硫氰酸盐的质量浓度在0.20~30.0 mg/L 范围内其色谱峰面积线性关系良好,相关系数分别为0.999 2、0.999 4,加标回收率分别为95.0%~101.1% 和95.5%~100.5%,5 次平行测定结果的相对标准偏差小于3%。该方法适用于尿液中草酸和硫氰酸盐的同时测定。 
Abstract:A method for simultaneous determination of oxalate and thiocyanate in urine by ion chromatography with suppressed conductivity detection was established. The chromatographic conditions were optimized by experiments. SH-AC-3 anion exchange column was used as separation column, 10.0 mmol/L Na2CO3 was used as eluent, and the flow rate was 1.0 mL/min, oxalic acid and thiocyanate could be completely separated from common anions such as chloride, phosphate and sulfate in urine by isocratic elution, and determined by suppressed conductivity detection. The peak area of oxalic acid and thiocyanate had good relationship with their mass concentrationin in the range of 0.20-30.0 mg/L with the correlation coefficient was 0.999 2 and 0.999 4, respectively. The recoveries oxalate and thiocyanate were 95.0%-101.1% and 95.5%-100.5%, respectively. The relative standard deviation of five parallel determinations results was less than 3%. The method is suitable for the simultaneous determination of oxalic acid and thiocyanate in urine.
