高效液相色谱法测定阿比特龙的纯度

摘要:建立测定阿比特龙纯度的高效液相色谱法。色谱条件如下:色谱柱为Agilent C18柱(250 mm×4.6 mm,3.5 μm),流动相为乙腈-水(1∶1),流量为1.2 mL/min,检测波长为254 nm,柱温为25℃,进样体积为20 μL。阿比特龙的质量浓度在40~520 μg/mL范围内与色谱峰面积线性关系良好,线性方程为A=31.957 2c+136.165 2,相关系数为0.999 7,检出限为0.24 μg/mL。样品测定结果的相对标准偏差为0.27%(n=6),标准样品回收率为99.12%~99.88%。该法专属性强,准确、可靠,具有较高的灵敏度和精密度。 
Abstract:A method of high performance liquid chromatography (HPLC) for the determination of abiraterone was established. The chromatographic conditions were as follows: Agilent C18 column (250 mm×4.6 mm, 3.5 μm), acetonitrile-water solution (1∶1) as the mobile phase with the flow rate of 1.2 mL/min, detection wavelength of 254 nm, column temperature of 25℃, and injection volume of 20 μL. The mass concentration of abiraterone was proportional to the chromatographic peak area in the range from 40 to 520 μg/mL. The linear regression equation was formulated as: A=31.957 2c +136.165 2 with the correlation coefficient of 0.999 7. The detection limit was 0.24 μg/mL. The relative standard deviation of the detection results was 0.27% (n=6). The recoveries of standard samples were in the range from 99.12% to 99.88%. The method is specific, accurate and reliable with high sensitivity and precision.
